‘Sorting-Out’ the Skeleton: The Role of Sorting Nexin 10 in Osteoclasts and Bone Disease
Purpose:
This proposal investigates the role of SNX10, a molecule that directs the transport of membrane proteins inside osteoclasts (OCs), giant bone-dissolving cells. Mutations in SNX10 cause autosomal recessive osteopetrosis (ARO), a rare but life-threatening bone disease in children associated with increased bone mass but increased risk of fractures and loss of the bone marrow cavity. This work will establish how SNX10 works in OCs and whether SNX10 can be targeted for the treatment of bone diseases.
